Overview

The CSB maintains a variety of instruments for studying the conformation and stability of macromolecules and for measuring the affinity and thermodynamics of biomolecular interactions. To set up core access and training, contact Noah Saracho. If you have additional questions or would like to set up a consultation, contact Heather Kroh.

Services and Fees

  • Find us in iLab
    • The Biophysical Instrumentation Facility and the Megalab Core have been combined to form the new ÌÀÍ·Ìõ CSB Macromolecular Biophysics Core in iLab. The Biomolecular Crystallography Facility was added in November 2024.
  • Training
    • Required of all first time users
    • Must be completed before self-service access is granted
    • Covers basic operation of the instrument
    • To schedule training*, contact Noah Saracho or Heather Kroh and provide an iLab billing number for you or your PI

  • Instrument Time
    • Includes access to instrument and accessories as scheduled by user
    • Short-term data file storage
    • Access to LAN and printer for report generation or file transfer
    • Basic operational assistance (initial set-up, instrument malfunction resolution)
    • Maintenance and repair of the instrument and accessories
  • Fees
    • Fees are set yearly by Core Management and subject to change
    • Current training fee for new users of CSB biophysical instruments is $59.80

    • Additional Charges: Damage to equipment caused by user negligence or abuse will result in charges for replacement or repair costs as well revocation of access privileges.

Core Instruments

  • Octet R8 Bio-layer Interferometer

    • Sartorius Octet R8
    • High-quality quantitative, kinetic, or affinity characterization of biomolecules
    • Can run up to 96 samples in a single experiment
    • Installed in January 2023
  • Mass Photometer

    • Reyfeyn TwoMP Mass Photometer
    • Measure molecular mass of proteins and
      protein assemblies in solution in their native state
    • Quick and accurate mass determination with minimal sample requirements (<20 µL, low nanomolar concentrations)
    • Installed in January 2023
  • Stopped-Flow Spectrometer

    • Kintek SF300X Stopped-flow Spectrometer
    • Measures changes in absorbance and/or fluorescence signals for kinetic and equilibrium titration experiments
    • Minimal first reaction time of 5 millisecond, with a sub-millisecond dead time
    • Computer-controlled servo motor (not air-driven)
    • Capacity for single- and double-mixing experiments
  • Circular Dichroism Spectropolarimeter

    • Chirascan VX CD spectrometer
    • User friendly data acquisition and analysis
    • Peltier temperature control

    • Determine binding affinities using fluorescently labeled ligands
    • Equipped with two LED lasers, blue and red

    • Verify protein quality, stability and reproducibility, with minimal volume needed (10 µL)
    • Measures intrinsic fluorescence of tryptophan and tyrosine residues
    • Can detect impurities, compare concentrations, and screen storage conditions

Accessing Data on the Network Drive

CD, fluorometer or calorimeter data saved to network J: drive can be accessed remotely.

  • Windows
    • Right click ‘Start’ and then click ‘Run’
    • °Õ²â±è±ðÌý°À°À³¦±ð²õ.¾±³Ù.±¹²¹²Ô»å±ð°ù²ú¾±±ô³Ù.±ð»å³Ü°À³¦²õ²ú²õ³ó²¹°ù±ð»å¾±²Ô²õ³Ù°ù
    • Click ‘OK’
    • It should prompt you to sign in. °Õ²â±è±ðÌý³Õ´¡±·¶Ù·¡¸éµþ±õ³¢°Õ°À²â´Ç³Ü°ù³Õ±«²Ô±ð³Ù±õ¶Ù
    • Enter your e-password.
    • Double click on the Data folder.
    • You should see folders for the CD, DSC, Fluoromax and ITC.
  • Mac
    • From the Finder menu, click ‘Go’ and then click ‘Connect to Server’
    • It should prompt you for the Server Address. Type smb://ces.it.vanderbilt.edu/csbsharedinstr
    • Click ‘Connect’
    • It should prompt you for your VUnetID and e-password.
    • Double click on the Data folder.
    • You should see folders for the CD, DSC, Fluoromax and ITC.
  • Linux
    • The easiest way to access it is through the file manager in Gnome:
    • Click ‘Places’ and then click ‘Connect to Server’

    • A window with the contents of your linux home dir may popup, but just close the popup and then click on the new icon for the share on the desktop. It will ask you for your e-password.
    • If you’re using KDE, then just use the file browser (konqueror or dolphin, e.g. with KDE v4+):
